Re-wiring the metabolism of cancer cells – therapeutically targeting glycolytic metabolism of blood cancers.
Acute myeloid leukaemia (AML) is a haematological malignancy in which the bone marrow accumulates abnormal myeloid cells. High levels of reactive oxygen species (ROS) have previously been shown to be produced by AML blast cells through NOX2. Using a limited panel of cell lines, ROS has previously been shown to promote AML blast proliferation by increasing glucose uptake as a consequence of overexpression of a critical glycolytic enzyme, 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase (PFKFB3). This relationship now needs to be substantiated in a larger of panel of AML cell lines and primary AML blasts.
